近日来自斯坦福大学的研究人员开发了一种新型的单细胞PCR微流体技术,并利用这一技术对数百个结肠癌细胞进行了单细胞基因表达分析,由此获得了人类结肠癌异质性图谱。相关研究成果发表在《自然生物技术》(Nature Biotechnology)杂志上。

肿瘤异质性是恶性肿瘤重要特征之一。肿瘤异质性可以表现在肿瘤分化水平及肿瘤功能水平上,出现异质性的抗原表达或出现不同生物特性细胞亚群。这种肿瘤异质性特性往往给肿瘤的研究和治疗带来极大的困难。

“在过去的数年里,我们一直致力于开发出一种单细胞基因组分析技术,”文章的资深作者、斯坦福大学工程学院及生物工程和应用物理系教授Stephen Quake说:“在新研究中我们利用开发的新技术获得了可喜的具有重要生物学意义的成果。”

研究人员利用这项新技术逐个地对细胞中的基因表达情况进行了分析,从中鉴别出了47种差异性表达的基因,并基于这些结果对结肠癌组织的细胞进行了亚型分类。

“在一次实验中,我们获得了特定组织细胞组成的‘快照’,鉴别出了不同的细胞亚型,并找到了识别这些细胞的新型标记物,”文章的第一作者Tomer Kalisky说。

新研究的重要发现之一就是揭示了肿瘤异质性产生的根源。长久以来科学家们认为肿瘤是在演进的过程中失去了基因组稳定,发生随机变化而生成了不同生物特性细胞亚群。新研究发现结肠癌细胞形成遵循了正常结肠细胞的分化过程。打个比喻,就如同漫画肖像,绘制的是同一张脸谱,只是某些特征被充分夸大了。癌细胞形成机制与正常细胞完全相同,只是小部分的细胞类型发生了不正常的扩大或减少。

“我们观察到结肠癌组织显示了与正常结肠上皮相似的谱系多样性,其中包括了不成熟组细胞样细胞和成熟的特化细胞,”Dalerba说。

此外,分析结果还显示在肿瘤组织中基因表达上越是接近不成熟细胞的细胞越是呈现恶性特征。基于这些结果,研究人员开发出了一种两基因分类系统,并证实相比于常规的病理学分级,这一简单的分析系统能够更准确地确定疾病的预后。

研究人员期望在未来能将这些技术广泛地应用到癌症的诊疗和预后分析中。
