Overview

Granulocyte-macrophagecolony-stimulatingfactor(GM-CSF)promotestheproliferationanddifferentiationofhematopoieticProgenitorcellsandthegenerationofneutrophils,eosinophils,andmacrophages.Insynergywithothercytokinessuchasstemcellfactor,IL-3,erythropoietin,andthrombopoietin,italsostimulateserythroidandmegakaryocyteprogenitorcells(Barredaetal.).GM-CSFisproducedbymultiplecelltypes,includingstromalcells,Panethcells,macrophages,dendriticcells(DCs),endothelialcells,smoothmusclecells,fibroblasts,chondrocytes,andTh1andTh17Tcells(Francisco-Cruzetal.).ThereceptorforGM-CSF(GM-CSFR)iscomposedoftwosubunits:thecytokine-specificαsubunit(GMRα;CD116)andthecommonsubunitβc(CD131)sharedwithIL-3andIL-5receptors(Broughtonetal.).GM-CSFRisexpressedonhematopoieticcells,includingprogenitorcellsandimmunecells,aswellasnon-hematopoieticcells.RecombinanthumanGM-CSF(rhGM-CSF)promotestheproductionofmyeloidcellsofthegranulocytic(neutrophils,eosinophilsandbasophils)andmonocyticlineagesinvivo.Ithasbeentestedformobilizationofhematopoieticprogenitorcellsandfortreatingchemotherapy-inducedneutropeniainpatients.GM-CSFisabletostimulatethedevelopmentofDCsthatingest,process,andpresentantigenstotheimmunesystem(Francisco-Cruzetal.).
