PreservetheActivityofSolubilizedMembraneProteins

ProteinPreservationDHPCpreservesthenativeconformationandthereforetheactivityofthesolubilizedproteins.Thethreedimensionalstructureandactivityofproteinsareretainednotonlyatthe10-15mMDHPCconcentrationsatwhichmaximumsolubilizationusuallyoccurs,butalsooveralargerangeofDHPCconcentrations(upto40mM).NotonlyisthethreedimensionalstructureandhencetheactivityofmostproteinretainedinexcessDHPC,buttheproteinsalsoappeartobestableinDHPC.SucharesultsuggeststhatthedirectinteractionofDHPCwithintegralmembraneproteinsisweak.IftheinteractionofDHPCwithmembraneproteinsisindeedweak,itfollowsthatintrinsicmembranelipidswillremainassociatedwiththemembraneproteins.

SolubilizationMechanismDHPCisthoughttoexertawedge-likeeffectontheneighboringlipids,mainlyduetoitsbulkypolargroupandshorthydrocarbonchains.ThisproducesmembranedestabilizationatrelativelylowDHPCconcentrations.AllmembranesinvestigatedaresolubilizedatanidenticalDHPC/lipidratio.ThisfindingistakenasevidencethatDHPCprimarilyinteractswiththelipidbilayerandnotwiththemembraneproteins.TheprincipleunderlyingthepreservationofthenativeproteinstructureistheinabilityofDHPCtodisplaceintrinsicmembranelipidsfromintegralmembraneproteins.
