TheHSP90α(C-Terminal)InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofHSP90αbindingtoitsproteintargetPPID(alsoknownasCyclophilinD).Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingHSP90α,PPID,andaninhibitorofchoiceisincubatedforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

OnlylimitedamountsofDMSO(<1%)canbeincludedasithasbeenshowntodisruptHSP90α:PPIDinteraction.GreenandbluedyesthatabsorblightintheAlphaScreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreen®assays.
