TheHSP90β(C-terminal)InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofHSP90βbindingtoitsproteintargetPPID(alsoknownasCyclophilinD).Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingHSP90β,PPID,andaninhibitorofchoiceisincubatedforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

AvoidDMSOconcentrationsabove0.5%.AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
