Background:

IFN-gammaisproducedmainlybyT-cellsandnaturalkillercellsactivatedbyantigens,mitogens,oralloantigens.ItisproducedbylymphocytesexpressingthesurfaceantigensCD4andCD8.IFNgammareceptorsareexpressedonalltypesofhumancellswiththeexceptionofmatureerythrocytes.IFN-gamma-receptorcomplexesarerapidlyinternalizedbyendocytosis.IFN-gammahasantiviralandantiparasiticactivitiesandalsoinhibitstheproliferationofanumberofnormalandtransformedcells.Interferon-gammasynergiseswithTNF-alphaandTNF-betaininhibitingtheproliferationofvariouscelltypes.ThegrowthinhibitoryactivitiesofIFN-gammaaremorepronouncedthanthoseoftheotherinterferons.However,themainBIOLOGicalactivityofIFN-gammaappearstobeimmunomodulatoryincontrasttotheotherinterferonsthataremainlyantiviral.InT-helpercells,IL-2inducesthesynthesisofIFN-gammaandothercytokines.IFN-gammaactssynergisticallywithIL-1andIL-2andappearstoberequiredfortheexpressionofIL-2receptorsonthecellsurfaceofT-lymphocytes.BlockingoftheIL-2receptorbyspecificantibodiesalsoinhibitsthesynthesisofIFN-gamma.
