ByWesternblotanalysisusingratbrainextractsofcortex,hypothalamus,midbrain,andhindbrain,theantibodyspecificallylabelsasingleband.Thepredictedmolecularweightofserotonin transporterbasedonitsnucleicacidsequenceis67,815. Theactualweightthatonemightobserve fromtissueextractscouldbehigherduetoglycosylation. WevalidatedtheantibodywithratSERToverexpressedinaninsectcellexpressionsystem. TheMWfromthatwasabout67kD. OurSERTantibodydilutionwas1:20,000usingchemiluminescentdetection.

Immunolabelingiscompletelyabolishedbypre-adsorptionwithsyntheticrat5HTtransporter(602-622).
