ImmunolabelingiscompletelyabolishedbypreadsorptionwithexcessGABA/BSAconjugate.
PhotoDescription:IHCimageofneuronsstainingforGABAintheratthalamus(above)andtheratcerebellum(belowleft,lowmagnification;belowright,highmagnification).Thetissuewasfixedwith4%formaldehydeinphosphatebuffer,beforebeingremovedandpreparedforvibratomesectioning.FloatingsectionswereincubatedatRTin10%goatseruminPBS,beforestandardIHCprocedure.Primaryantibodywasincubatedat1:20000for48hours,goatanti-rabbitsecondarywassubsequentlyaddedfor1hourafterwashingwithPBS.Lightmicroscopystainingwasachievedwithstandardbiotin-streptavidin/HRPprocedureandDABchromogenandhemotoxylincounterstainintheratthalamus.
