Thereareatleast20structurallyandfunctionallyrelatedBMPs,whicharemembersoftheTGF-betasuperfamily.BMPswereoriginallyidentifiedasproteinregulatorsofcartilageandboneformation.Theyarealsoinvolvedinembryogenesisandmorphogenesisofvarioustissuesandorgans.BMPsregulatethegrowth,differentiation,chemotaxisandapoptosisofvariouscelltypes,includingmesenchymalcells,epithelialcells,hematopoieticcellsandneuronalcells.SimilartootherTGF-betafamilyproteins,BMPsarehighlyconservedacrossanimalspecies.

BMP4isimplicatedinfibrodysplasiaossificansprogressive.Itisreportedtobeuniquelyoverexpressedinlymphoblastoidcellsandpreosseousfibroproliferativelesions.BMPs1to6arealsoreportedtobeexpressedinprostaticadenocarcinomaswithBMPs1to5alsoexpressedinbothbenignandprostatichyperplasiaandocularmelanoma.

Smad1translocationtothenucleusisobservedaftertheadditionofBMP4(alsodesignatedBMP2B),suggestingthatBMP4mayplayaroleinactivationoftheSmadpathway.BMPissecretedintotheextracellularmatrix.
