BACKGROUND

Thymidinekinase(TK)belongstoagroupofenzymessuchasdihydrofolatereductase,thymidylatesynthase,andDNApolymerasethatareinvolvedinDNAsynthesisandprecursorproduction.Highlevelsoftheseenzymesarepresentinproliferatingcells,andlowlevelsarefoundinrestingcells,duetomultipleregulatorymechanismsthatensureexclusiveexpressionoftheseenzymesinreplicatingcells.TKisresponsIBLeforcatalyzingthephosphorylationofthymidine,whichfunctionsasapartofthepyrimidinesalvagepathwayinvolvedinDNAsynthesis.TheactivitiesofenzymessuchasTKareessentialfortheactivationofseveralchemotherapeuticallyimportantnucleosideanaloguesthatareadmiNISTeredasprodrugs.
