TheInsulin-likegrowthfactors(IGFs)aremitogenicpolypeptidegrowthfactorsthatstimulatetheproliferationandsurvivalofvariouscelltypes,includingmuscle,boneandcartilagetissueinvitro.Theyarepredominantlyproducedintheliver.IGFsbelongtotheInsulingenefamilyandaresimilarbystructureandfunctiontoinsulin,buthaveamuchhighergrowth-promotingactivity.IGF1expressionisregulatedbygrowthhormone,whileIGF2expressionisinfluencedbyplacentalactogen.BothIGF1andIGF2signalthroughthetyrosinekinasetypeIreceptor(IGFIR).MatureIGFsaregeneratedthroughproteolyticprocessingofinactiveprecursorproteins.RecombinanthumanIGF1isaglobularproteincontaining70aminoacids,and3intra-moleculardisulfidebonds.
