  • 1.Allreagentswerebroughtuptoroomtemperaturefor30minutespriortouse.The1xWashBufferwaspreparedbyadding20mLof25xWashBufferConcentrateto480mLofdistilled/deionizedwaterandmixingthoroughly.
  • 2.ThevialofStandardwasreconstitutedwith1mLofSampleDiluent,mixed,andallowedtositfor15minuteswithgentleagitation.
  • 3.Thestandardcurvewaspreparedbycreatinga2-folddilutionseriesofsevenstandards(includingtheoriginalundilutedvial)usingSampleDiluent.SampleDiluentaloneservedasthe0pg/mLstandard.
  • 4.Theassayplatewasremovedfromthefoilpouchand100μLofeachstandardandsamplewereaddedtotheappropriatewells,intriplicate.Theplatewascoveredwiththeadhesivestripprovidedandincubatedfor2hoursat37°C.
  • 5.Approximately10minutesbeforetheincubationended,a1xBiotin-antibodysolutionwaspreparedbydiluting60μLof100xBiotin-antibodyinto5940μLofBiotin-antibodyDiluent.

  • 9.Eachwellwasaspiratedandwashed,repeatingtheprocesstwotimesforatotalofthreewashes.Eachwellwaswashedbyfillingeachwellwith1xWashBufferandlettingitstandfor2minutes.Afterthelastwash,remainingWashBufferwasremovedandtheplatewasinvertedandblottedagainstclean,absorbentpapertowels.
  • 10.100μLof1xHRP-avidinsolutionwasaddedtoeachwell,theplatewascoveredwithanewadhesivestrip,andincubatedfor1hourat37°C.
  • 11.Theaspiration/washprocedurefromStep9wasrepeatedforanadditional5washes.
  • 12.90μLofTMBSubstratewasaddedtoeachwell.Theplatewasprotectedfromlightandincubatedfor15-30minutesat37°C,withperiodiccheckingtopreventoverdevelopment.
  • 13.50μLofStopSolutionwasaddedtoeachwellandmixedthoroughly.Theopticaldensity(OD)ofeachwellwasmeasuredwithin5minutesusingamicroplatereadersetto450nm.
  • 14.AstandardcurvewasgeneratedbyplottingtheODvalueforeachstandardonthey-axisagainsttheconcentrationonthex-axis.AlineofbestfitthroughthepointsonthegraphwasusedtogenerateanequationtocalculateCXCL16concentrationsofthesamplesbasedontheiraverageODvalues.
ExperimentalNotesTheTMBsubstrate(Lot03190614)usedforthisassaywaslightbluepriortoaddition.The1:100dilutionofthepositivecontrol(humanserum)reportedmuchhighervaluesthanitslessdilutecounterpartsandshouldbetreatedasoutliers.
