Description
ThethreemammalianisoformsofTGF-β,TGF-β1,β2,andβ3,signalthroughthesamereceptorandelicitsimilarBIOLOGicalresponses.Theyaremultifunctionalcytokinesthatregulatecellproliferation,growth,differentiationandmotility,aswellassynthesisanddepositionoftheextracellularmatrix.Theyareinvolvedinvariousphysiologicalprocesses,includingembryogenesis,tissueremodelingandwoundhealing.Theyaresecretedpredominantlyaslatentcomplexes,whicharestoredatthecellsurfaceandintheextracellularmatrix.ThereleaseofthebiologicallyactiveTGF-βisoformfromalatentcomplexinvolvesproteolyticprocessingofthecomplexand/orinductionofconformationalchangesbyproteinssuchasthrombospondin-1. TGF-β2hasbeenshowntoexertsuppressiveeffectsonIL-2-dependentT-cellgrowth,andmayalsohaveanautocrinefunctioninenhancingtumorgrowthbysuppressingimmuno-surveillanceoftumordevelopment.RecombinantHumanTGF-β2isa25.0 kDaproteincomposedoftwoidentical112 aminoacidpolypeptidechainslinkedbyasingledisulfide bond.
