Scientific DescriptionUbiquitin (Ub) was first identified in 1975 as a 76 amino acid protein that is highly conserved in all eukaryotic cells. Ubiquitin requires three-types of enzyme to conjugate to other proteins: Ub-activating enzyme (E1), Ub-conjugating enzymes (E2), and Ub-ligases (E3). Ubiquitin can be covalently attached to lysine (Lys) residues of target proteins either as a monomer (monoubiquitin), a polymer (polyubiquitin chains) or a linear polymer (linear polyubiquitin chains). All seven Lys residues (Lys6, Lys11, Lys27, Lys29, Lys33, Lys48, and Lys63) can be ubiquitinated. The ubiquitinated peptides have different functions depending on the Lys residue of the ubiquitin that is linked: Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in lysosomal degradation; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, and DNA-damage responses.
Storage & StabilityThe antibody is delivered in lyophilized powder. Upon receipt, please centrifuge the antibody at 12,000 x g for 20 seconds, and then reconstitute it with the antibody stabilizer provided. The stabilizer contains PBS, 50% glycerol, and 0.01% sodium azide. Please store the antibody at -20°C. Avoid repeated freeze/thaw cycles. Antibody is stable for 12 months from date of reconstitution. Equilibrate the antibody to room temperature for 2 minutes and mix by gently pipetting before use.**Research purposes only.
