CD152[CTLA-4]andCD28,togetherwiththeirligandsB7-1andB7-2,constituteoneofthedominantcostimulatorypathwaysthatregulateTandBcellresponses.CD152andCD28arestructurallyhomologousmoleculesthataremembersoftheimmunoglobulin(Ig)genesuperfamily.BothCD152andCD28arecomposedofasingleIgV-likeextracellulardomain,atransmembranedomainandanintracellulardomain.CD152andCD28arebothexpressedonthecellsurfaceasdisulfide-linkedhomodimersorasmonomers.CD152wasoriginallyidentifiedasagenethatwasspecificallyexpressedbycytotoxicTlymphocytes.However,CD152transcriptshavesincebeenfoundinbothTh1andTh2,andCD4⁺andCD8⁺Tcellclones.Whereas,CD28expressionisconstitutiveonthesurfacesof95%ofCD4⁺Tcellsand50%ofCD8⁺TcellsandisdownregulateduponTcellactivation,CD152expressionisupregulatedrapidlyfollowingTcellactivationandpeaksapproximately24hoursfollowingactivation.AlthoughbothCD152andCD28canbindtothesameligands,CD152bindstoB71andB72with20-100-foldhigheraffinitythanCD28.
