Abstract

Metabolisminmammalsisregulatedbycomplexinterplayamongdifferentorgans.Fattyacidsynthesisisincreasedinwhiteadiposetissue(WAT)whenitisinhibitedintheliver.Hereweidentifyglycoproteinnon-metastaticmelanomaproteinB(Gpnmb)asoneliver鈥揥ATcross-talkfactorinvolvedinlipogenesis.Inhibitionofthehepaticsterolregulatoryelement-bindingproteinpathwayleadstoincreasedtranscriptionofGpnmbandpromotesprocessingofthemembraneproteintoasecretedform.GpnmbstimulateslipogenesisinWATandexacerbatesdiet-inducedobesityandinsulinresistance.Inhumans,Gpnmbistightlyassociatedwithbodymassindexandisastrongriskfactorforobesity.Gpnmbinhibitionbyaneutralizingantibodyorliver-specificknockdownimprovesmetabolicparameters,includingweightgainreductionandincreasedinsulinsensitivity,probablybypromotingthebeigingofWAT.TheseresultssuggestthatGpnmbisaliver-secretedfactorregulatinglipogenesisinWAT,andthatGpnmbinhibitionmayprovideatherapeuticstrategyinobesityanddiabetes.
