IntendedUseTheToxoplasmagondiiIgGaviditytestisintendedtodifferentiatebetweenpastandacuteinfection.
IntroductionTheaviditydeterminationisadiagnosticmethodwhichisusedtodifferentiatearecent(acute)andamoredistant(past)infectionwithToxoplasmagondiiinpatientsera.Avidityisthebindingforceoftheantibody(serumspecimen)withthecorrespondingantigen.
LowavidIgGantibodiesintheearlystageofinfectioncanbedifferentiatedfromhighavidantibodiesassociatedwithapastinfection.
ThedeterminationofIgGantibodyavidityisanadditionalanalysistotheclassicSEROlogyinregardtothestatusofaToxoplasmagondiiinfection.
PrinciplesoftheassayMicrotiterstripwellscoatedwithToxoplasmaantigenareincubatedwithdilutedserumspecimen(dualpipetting).Afterwashingonewellisincubatedwithavidityreagentandthecorrespondingwellwithwashingbuffer.Inthisstepthelowavidityantibodiesareremovedfromtheantigenswhereasthehighavidityonesarestillboundtothespecificantigens.
AntihumanIgGlabelledwithperoxidaseisadded.TheimmuncomplexisvisualizedwithTMBtogiveabluereactionproduct.Stopsolutionisaddedtostopthereactionandchangingthecolourofthereactionproductintoyellow.Absorbanceat450nmisreadusinganELISAmicrowellplatereader.
StorageandStABIlityThereagentsarestableuptotheexpirydatestatedonthelabelwhenstoredat2-8°C.
Afterfirstopeningthereagentsarestableuptotheexpirydate.
LimitationsoftheTestBacterialcontaminationorrepeatedfreeze-thawcyclesofthespecimenmayaffecttheabsorbancevalues.Diagnosisofaninfectiousdiseaseshouldnotbeestablishedonthebasisofasingletestresult.Aprecisediagnosisshouldtakeintoconsiderationclinicalhistory,symptomatologyaswellasserologicaldata.
Inimmunocompromizedpatientsandnewbornsserologicaldataonlyhaverestrictedvalue.
Aresultofhighaviditycannotexcludethepossibilityofarecentinfection.
