IntendedUse:TheCMVIgMELISAisintendedforuseinthedetectionofIgMantibodiestoCytomegalovirus(CMV)infectioninhumanserum.

Introduction:CytomegalovirusisaherpesvirusandaleADIngBIOLOGicalfactorcausingcongenitalabnormalitiesandcomplicationsamongthosewhoreceivemassivebloodtransfusionsandimmunosuppressivetherapy.Abouthalfthepregnantwomenwhocontractaprimaryinfectionspreadthediseasetotheirfetus.Whenacquiredin-utero,theinfectionmaycausementalretardation,blindness,and/ordeafness.
SEROlogicaltestsfordetectingthepresenceofantibodytoCMVcandiagnoseactiveorrecentinfectionandprovidevaluableinformationregardingthehistoryofpreviousinfection.Thesetestsarealsousefulinscreeningbloodfortransfusionsinnewbornsandimmuno-compromisedrecipients.TheCMVIgMELISAisanaccurateserologicmethodtodetectCMVIgMantibodyforidentificationofCMVinfection.

Principleofthetest:PurifiedCMVantigeniscoatedonthesurfaceofmicrowells.Dilutedserumisaddedtothewells,andtheCMVIgMspecificantibody,ifpresent,bindstotheantigen.Allunboundmaterialsarewashedaway.HRP-conjugateisadded,whichbindstotheantibody-antigencomplex.ExcessHRP-conjugateiswashedoffandasolutionofTMBReagentisadded.Theenzymeconjugatecatalyticreactionisstoppedataspecifictime.TheintensityofthecolorgeneratedisproportionaltotheamountofIgMspecific-antibodyinthesample.Theresultsarereadbyamicrowellreadercomparedinaparallelmannerwithcalibratorandcontrols.

LimitationoftheTest:1.ReliableandreproducIBLeresultswillbeobtainedwhentheassayprocedureiscarriedoutwithacompleteunderstandingofthepackageinsertinstructionsandwithadherencetogoodlaboratorypractice.
2.Thewashprocedureiscritical.Insufficientwashingwillresultinpoorprecisionandfalselyelevatedabsorbancereadings.
3.Serumsamplesdemonstratinggrosslipemia,grosshemolysis,orturbidityshouldnotbeusedwiththistest.
