ShortDescription:InthisassaytheCERULOPLASMINpresentinserumsamplereactswiththeanti-CERULOPLASMINantibodieswhichhavebeenadsorbedtothesurfaceofpolystyrenemicrotitrewells.Aftertheremovalofunboundserumproteinsbywashing,anti-CPantibodiesconjugatedwithhorserADIshperoxidase(HRP),areadded.Theseenzyme-labeledantibodiesformcomplexeswiththepreviouslyboundserumCP.Followinganotherwashingstep,theenzymeboundtotheimmunosorbentisassayedbytheadditionofachromogenicsubstrate,3,3’,5,5’-tetramethylbenzidine(TMB).ThequantityofboundenzymevariesdirectlywiththeconcentrationofCPinthesampletested;thus,theabsorbance,at450nm,isameasureoftheconcentrationofCerinthetestsample.ThequantityofCPinthetestsamplecanbeinterpolatedfromthestandardcurveconstructedfromthestandards,andcorrectedforserumdilution.
