ShortDescription:InthisassaytheIgMpresentinsamplesreactswiththeanti-IgMantibodieswhichhavebeenadsorbedtothesurfaceofpolystyrenemicrotitrewells.Aftertheremovalofunboundproteinsbywashing,anti-IgMantibodiesconjugatedwithhorserADIshperoxidase(HRP),areadded.Theseenzyme-labeledantibodiesformcomplexeswiththepreviouslyboundIgM.Followinganotherwashingstep,theenzymeboundtotheimmunosorbentisassayedbytheadditionofachromogenicsubstrate,3,3’,5,5’-tetramethylbenzidine(TMB).ThequantityofboundenzymevariesdirectlywiththeconcentrationofIgMinthesampletested;thus,theabsorbance,at450nm,isameasureoftheconcentrationofIgMinthetestsample.ThequantityofIgMinthetestsamplecanbeinterpolatedfromthestandardcurveconstructedfromthestandards,andcorrectedforsampledilution.
