Background

Thisantibodyisdesigned,produced,andvalidatedaspartofacollaborationbetweenRocklandandtheNationalCancerInstitute(NCI).AGAP2(alsoknownasArf-GAPwithGTPase,ANKrepeatandPHdomain-containingprotein2)isaGTPaseactivatingproteinthatinactivatesArf.TheexpressionofAGAP2isamplifiedinhumanglioblastomacells.GTPase-activatingprotein(GAP)forARF1andARF5,whichalsoshowsstrongGTPaseactivity.Itmediatesanti-apoptoticeffectsofnervegrowthfactorbyactivatingnuclearphosphoinositide3-kinase.Isoform1participatesinthepreventionofneuronalapoptosisbyenhancingPI3kinaseactivity.Itaidsthecouplingofmetabotropicglutamatereceptor1(GRM1)tocytoplasmicPI3kinasebyinteractingwithHomerscaffoldingproteins,andseemstomediateanti-apoptoticeffectsofNGFbyactivatingnuclearPI3kinase.Isoform2doesnotstimulatePI3kinasebutmayprotectcellsfromapoptosisbystimulatingAkt.Italsoregulatestheadapterprotein1(AP-1)-dependenttraffickingofproteinsintheendosomalsystem.Itseemstobeoncogenic.Itisoverexpressedincancercells,preventsapoptosisandpromotescancercellinvasion.Anti-AGAP2AntibodyisusefulforresearchersinterestedinDevelopmentalBIOLOGy,apoptosis,Cancer,Immunology,andNuclearSignalingresearch.
