TheclinicaluseofthepotentantitumoractivityofTNF-αhasbeenlimitedbytheproinflammatorysideeffectsincludingfever,dose-limitinghypotension,hepatotoxicity,intravascularthrombosis,andhemorrhage.DesigningclinicallyapplicableTNF-αmutantswithlowsystemictoxicityhasbeenanintensepharmacologicalinterest.HumanTNF-α,whichbindstothemurineTNF-α55butnottothemurineTNF-α75,exhibitsretainedantitumoractivityandreducedsystemictoxicityinmicecomparedwithmurineTNF-α,whichbindstobothmurineTNFreceptors.Basedontheseresults,manyTNF-αmutantsthatselectivelybindtoTNF-α55havebeendesigned.Thesemutantsdisplayedcytotoxicactivitiesontumorcelllinesinvitro,andexhibitedlowersystemictoxicityinvivo.
