Abstract
TheaimofthepresentstudyistoinvestigatethemoleculareffectsofGoldnanoparticles(GNPs)onthehumandermalfibroblasts-fetal(HDF-f)atthelevelofproteinexpression.20-nmGNPswerepreparedusingchemicalreductionmethod.AfterHDF-fweretreatedwith200microMGNPsfor1,4and8h,proteinexpressionprofileswereobtainedusingtwo-dimensionaldifferencegelelectrophoresis(2D-DIGE)andmassspectra(MS)analysis.Theobtaineddifferentialexpressedproteinswereanalyzedbyclustering,genemicroarraypathwayprofiler(GenMAPP)andIngenuitypathwayanalysis(IPA)analysis,andverifiedbywesternblot.40proteinspotswerefilteredwithdifferentexpressioninabundanceinallthree-cultureperiodsand24uniqueproteinswereidentified.BioinformaticsanalysisresultsindicatedthatGNPsmighthaveaninfluenceontheHDF-fintheaspectsofsignaltransduction,actinCytoskeleton,energymetabolism,oxidativestresscelltranscriptionfactor,etc.ComparedwiththegeneexpressioneffectsinducedbyGNPsinourpreviousresearch,certainrelationshipsatmolecularlevelafterHDF-ftreatedwithGNPswereidentified.TheproteomicanalysisusedherewouldalsobeausefultooltoimprovethemechaNISTicunderstandingofnanomaterialsbiocompatibility.
