Thedevelopmentoftherapeuticmethodsthatcaneffectivelydelaytumorgrowth,inhibittumormetastases,andprotectthehostfromtumorrecurrencestillfaceschallenges.Nanoparticle-basedcombinationtherapymayprovideaneffectivetherapeuticstrategy.Herein,weshowthatbovineserumalbumin(BSA)-bioinspiredgoldnanorods(GNRs)wereloadedwithanimmunoadjuvantforcombinedphotothermaltherapy(PTT)andimmunotherapyforthetreatmentofmelanoma.Inthiswork,cetyltrimethylammoniumbromide(CTAB)-coatedGNRsweresuccessivelydecoratedwithpolyethyleneglycol(PEG)andBSA,andloadedwithanimmunoadjuvantimiquimod(R837).ThesynthesizedmPEG-GNRs@BSA/R837nanocomplexesundernear-infrared(NIR)irradiationcouldeffectivelykilltumorsandtriggerstrongimmuneresponsesintreatingmetastaticmelanomainmice.Furthermore,thenanocomplex-basedPTTpreventedlungmetastasisandinducedastronglong-termantitumorimmunitytoprotectthetreatedmicefromtumorrecurrence.Thenanocomplex-basedPTTincombinationwithimmunotherapymaybepotentiallyemployedasaneffectivestrategyforthetreatmentofmelanomaandothermetastaticcancers.
