Myelin-oligodendrocyteglycoprotein(MOG)isamemberofthe immunoglobulin(Ig)superfamily,exclusivelyexpressedinthecentral nervoussystem(CNS).MOGisanintrinsicmembraneprotein characterizedbyaN-terminalextracellularimmunoglobulin-like variable(Ig-likeV-type)domain,twohydrophobictransmembrane
domainsandacytoplasmicC-terminalregion.TheN-terminalMOG domainhasstronghomologywiththeN-terminusofbutyrophilin,a proteinexpressedinthelactatingmammarygland.HumanMOGgene islocalizedtochromosome6p22-p21.3(bandCofmouse chromosome17)atthedistalendoftheMHCclassIbregion.Despite
thesimilarnames,oligodendrocyte-myelinglycoprotein(OMG)is aseparateproteinencodedwithinalargeintronoftheNF1gene. The2glycoproteinsareassociatedspecificallywitholigodendrocytes andmyelin,buthavequitedifferentrolesinmyelinogenesisandare structurallyunrelated.MOGisanintrinsicmembranemoleculewith2 transmembranedomains,whereasOMGisanchoredintheouter leafletoftheplasmamembranethroughaglycophospholipidtail.OMG belongstothefamilyofproteinswithaseriesoftandemleucine-rich
repeats,whileMOGisamemberoftheIgsuperfamily.
MOGcontainsnineexonsandeightseparatingintrons,givingrise toatleasteightalternativelysplicedvariantsencodingforthe MOG-alpha1-4andMOG-beta1-4isoforms(16-26kDa).The differentMOGisoformsmayinteracttoformhomo-and heterodimersandtrimers(55and78kDa).Duringthelaststepof myelinogenesis,MOGisexpressedintheCNSontheoutermost surface(externallamella)ofmaturemyelinsheathsandonthecell surfaceofmyelinatingoligodendrocytes.MOGhasavarietyof functionsincludingaroleasacellularadhesionmolecule.Itmay beinvolvedalsointhecompletionand/ormaintenanceofthe myelinsheathandincell-cellcommunication.MOGisalsothought tofunctionasaregulatorofoligodendrocytemicrotubulest
ABIlity andasamediatorofinteractionsbetweenmyelinandtheimmune systeminthecomplementcascade.AlthoughMOGisarelatively minorcomponentofthemyelinmembrane,itisaprimaryautoantigen targetinvolvedinthepathogenesisofimmune-mediated demyelinatingdiseasesincludingexperimentalautoimmune
encephalomyelitis(EAE)andmultiplesclerosis. TheMOG35-55peptideisanimmunodominantepitopeofMOG
thatinducesstrongTandBcellresponses.Asingleinjectionof thispeptidefragmentcanproduceanexacerbating-remitting neurologicdiseasewithextensiveplaque-likedemyelination,which mayserveasamodelforinvestigatingmultiplesclerosis.

RatandmouseMOG35-55peptideis100%conservedcommon marmoset,95%inchimp,bovine,Pongopygmaeus(Orangutan),pig,andhumanMOG.MOG35-55sequenceisfoundinMOG isoformsalpha-1,alpha-2,beta-2,beta-3,beta-4,andisoforms9.
