TheJMJD3HomogeneousAssayKitisdesignedtomeasureactivityoftheJMJD3forscreeningandprofilingapplications,usingAlphaLISA^®technology.ThekeytotheJMJD3HomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,asamplecontainingJMJD3enzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:

AvoidgreenandbluedyesthatabsorblightintheAlphascreen^®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN₃)ormetalions(Fe²⁺,Fe³⁺,Cu²⁺,Zn²⁺andNi²⁺).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen^®assays.
