HumanT-cellimmunoreceptorwithIgandITIMdomains(TIGIT)isareceptorthatisexpressedonthesurfaceofhumanTcellsandNKcellsthatbindstoCD155andCD112onthesurfaceofdendriticcells.BindingofTIGITwithCD155orCD112resultsininhibitionofTcellandNKcellactivation.Antibodiesandotheragentsthatinhibitthissignalingpathwayhavebeenshowntoincreasetheimmuneresponse,especiallyinthecaseofcertaincancers.

Description:

TheTIGIT:CD112HomogeneousAssayKitisdesignedtomeasuretheinhibitionofhumanT-cellimmunoreceptorwithIgandITIMdomains(TIGIT)bindingtoCD112(PVRL2/Nectin-2).TheTIGIT:CD112HomogeneousAssayKitcomesinaconvenientAlphaLISA®formatwithpurifiedbiotinylatedTIGIT,His-taggedCD112,andassaybuffertoperformatotalof384reactions.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingTIGITandaninhibitorofchoiceisincubatedwiththeCD112for60minutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreadingtheAlpha-counts.

Contraindications:

OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptTIGIT:CD112interaction.AvoidgreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
