TheBRD2(BD2)InhibitorScreeningAssayKitisdesignedtomeasuretheinhibitionofBRD2bromodomain2(BD2)frombindingtoitssubstrate.ThekeytotheBRD2(BD2)InhibitorScreeningAssayKitisthehighlyspecificbindingoftheBRD2bromodomain2totheacetylatedhistonesubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingBRD2bromodomain2andaninhibitorofchoiceisincubatedwiththebiotinylatedsubstrateforthirtyminutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreadingtheAlpha-counts.

Contraindications:

DMSOabove0.5%.OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptBRD-ligandinteraction.AvoidgreenandbluedyesthatabsorblightintheAlphascreen^®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN₃)ormetalions(Fe²⁺,Fe³⁺,Cu²⁺,Zn²⁺andNi²⁺).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen^®assays.
