TheBasicHRTargetingVectorforGeneKnock-In/Out(MCS1-LoxP-MCS2-MCS3-pA-LoxP-MCS4)featuresfourdifferentMCSs—cloneyourhomologyarmsintoMCS1andMCS4,yourexpressioncassetteintoMCS3,andanyadditionalsequencessuchassmallRNAsthatdon’tneedapolyAtailintoMCS2—aswellastwoLoxPsitesthatcanbeusedtoremovetheexpressioncassetteafteritisnolongerneeded(learnmoreaboutCre-LoxPexcisionhere).

WhyuseanHRtargetingvector?

Eventhoughgeneknock-outscanresultfromDSBscausedbyCas9alone,SBIrecommendstheuseofHRtargetingvectors(alsocalledHRdonorvectors)formoreefficientandprecisemutation.HRdonorscansupplyelementsforpositiveornegativeselectionensuringeasieridentificationofsuccessfulmutationevents.Inaddition,HRdonorscanincludeupto6-8kbofopenreadingframeforgeneknock-insortagging,and,whensmallmutationsareincludedineither5’or3’homologyarms,canmakespecific,targetedgeneedits.
