Capan-1 Cell Line:

Pancreatic cancer remains one of the deadliest tumors with a poor prognosis and life expectancy. According to the National Pancreatic Cancer Foundation (NPCF), only 5% of pancreatic cancer patients will be still alive five years after the diagnosis. The Capan-1 cell line was derived from a 40-year-old male patient of pancreatic cancer. These tumorigenic cells exhibit an epithelial cell morphology and make suitable transfection hosts. Capan-1 cells have been known to produce gastric-type mucin, and express CFTR (cystic fibrosis transmembrane conductance regulator). The Capan-1 pancreatic cell line has proven to be a reliable host for transfection studies that aid investigative research for new drugs. In addition to being a suitable host for pancreatic cancer studies, this cell line is helpful in molecular biology as well as in drug discovery research. Cyclophilin B silencing efficiency was determined by qRT-PCR in the Capan-1 cells transfected by Cyclophilin B siRNA or non-silencing siRNA control following the recommended transfection protocol. Cyclophilin mRNA expression levels were measured 48 hours post-transfection. 18S rRNA levels were used to normalize the Cyclophilin B data. Values are normalized to untreated sample. Data are presented as means ± SD (n=6).

DNA plasmid expressing Cyclophilin B or siRNA targeting Cyclophilin B were transfected into Capan-1 cells following Altogen Biosystems transfection protocol. At 72 hours post-transfection the cells were analyzed by Western Blot for protein expression levels (normalized by total protein, 10 µg of total protein loaded per each well). Untreated cells used as a negative control.
