  • StockSolutionofGlucose
  1. EnsurethattheinvitromodificationofDNAprotocolforLactobacillusplantarumhasbeenfollowed.
  2. Thaw50ulofcompetentcellsonicefromaliquotsin-80°Cfreezer.
  3. Addbetween1ngand3ugofplasmidDNAin5ulofTEbufferto50uloffreshlypreparedcompetentcellsandtransfertoprechilledcuvetteforelectroporation(interelectrodedistance1mm).
  4. Electroporationdoneat13Kvcm-1(timeconstant:2-4ms),Setelectroporationmachineto25μFand200Ω.
  5. Add500ulofMRSmediacontaining80mMMgCl2and55mMGlucose
  6. Incubatefor2hoursat30°C.
  7. PlatecellsonMRSplates+antibioticresistance.
