Endopeptidasecleavagegeneratesafunctionallydistinctisoformof CTRP12 withanalteredoligomericstateandsignalingspecificity

Adiposetissue-derivedadipokinesareanimportantclassofsecretedmetabolicregulatorsthatmediatetissuecrosstalktocontrolsystemicenergybalance.WerecentlydescribedC1q/TNF-relatedprotein-12(CTRP12),anovelinsulin-sensitizingadipokinethatregulatesglucosemetabolisminliverandadiposetissue.However,thebiochemicalpropertiesof CTRP12 anditsnaturallyoccurringcleavedisoformhavenotbeencharacterized.Here,weshowthat CTRP12 isasecretedhormonesubjectedtomultiplefunctionallyrelevantposttranslationalmodificationsathighlyconservedresidues.Forexample,Asn-39isglycosylated,whileCys-85mediatestheassemblyofhigher-orderoligomericstructure.EndopeptidasecleavageatLys-91generatesacleavedglobulargCTRP12isoform,theexpressionofwhichisincreasedbyinsulin.PCSK3/furinwasidentifiedasthemajorproproteinconvertaseexpressedbyadipocytesthatmediatestheendogenouscleavageof CTRP12.CleavageatLys-91iscontextdependent:mutationofthechargedArg-93toAlaontheP2positionenhancedcleavage,andtriplemutations(K90A,K91A,R93A)abolishedcleavage.Importantly,thetwoisoformsof CTRP12 differinoligomericstructuresandarefunctionallydistinct.Thefull-lengthproteinformstrimersandlargercomplexesandthecleavedisoformconsistedofpredominantlydimers.Whilefull-lengthfCTRP12stronglyactivatedAktsignalinginH4IIEhepatocytesand3T3-L1adipocytes,gCTRP12preferentiallyactivatedMAPkinase(Erk1/2andp38-MAPK)signaling.Further,onlyfCTRP12improvedinsulin-stimulatedglucoseuptakeinadipocytes.Theseresultsrevealanovelmechanismcontrollingsignalingspecificityandfunctionofahormoneviacleavage-dependentalterationinoligomericstate.WeiZ, LeiX, SeldinM, WongGW..JBiolChem. 2012Aug31.[Epubaheadofprint]
