SolubleCD204/MSR1/SR-A ELISAKitCD204MacrophageScavengerReceptorTypesI (MSR1)SR-A(ScavengerreceptorclassAmember1)Stromalmacrophageexpressing CD204 isassociatedwithtumoraggressivenessinlungadenocarcinomaBACKGROUND:Tumortissueiscomposedofvariablenumbersofcancercellsandstromalcells,andtumor-associatedmacrophagesarerecruitedintocancer-inducedstromaandproduceaspecificmicroenvironment.Alternatively,activatedmacrophages(M2phenotype)areknowntoberelatedtotumorprogressionandoutcome,and CD204 hasbeenreportedtobeexpressedinM2macrophagesinsometumors.
METHODS:Toinvestigatewhether CD204-positivemacrophagesreflecttumoraggressivenessinadenocarcinomaofthelung,weinvestigatedtherelationshipsbetweenthenumbersof CD204-positivestromalmacrophagesandbothclinicopathologicalfeaturesandoutcomein170consecutiveresectedcases.Wealsoexaminedtherelationshipsbetweenthenumbersof CD204-positivemacrophagesandtheexpressionlevelsofcytokinesinvolvedinthemigrationanddifferentiationofM2macrophages.
RESULTS:Thenumbersof CD204-positivemacrophagesweresignificantlycorrelatedwithseveralprognosticfactors.Thelog-ranktestshowedasignificantassociationbetweenthenumbersof CD204-positivemacrophagesandapooroutcome(p=0.0073),whereasthenumbersofmacrophagesexpressingCD68,apan-macrophage/monocyte
Marker,wereofmarginalprognosticsignificance(p=0.0789).WeevaluatedassociationsbetweenthelevelsofexpressionofthecytokinesIL-6,IL-10,IL-12a,IL-12b,M-colony-stimulatingfactor,IFN-gamma-.,andmonocytechemoattractantprotein-1incancertissueandthenumbersof CD204-positivemacrophages.TheexpressionlevelsofIL-10andmonocytechemoattractantprotein-1,whichareinvolvedindifferentiation,accumulation,andmigrationofM2macrophages,weresignificantlycorrelatedwiththenumbersof CD204-positivemacrophages(p=0.031andp=0.031,respectively).
CONCLUSION:Thesefindingsdemonstratedthat CD204-positivemacrophagesclearlyreflectthetumor-promotingphenotypeoftumor-associatedmacrophagesinlungadenocarcinoma.OhtakiYetal.JThoracOncol. 2010Oct;5(10):1507-15.OverexpressionofCD163, CD204 andCD206onalveolarmacrophagesinthelungsofpatientswithseverechronicobstructivepulmonarydiseaseWehavepreviouslyreportedthatthelungsofpatientswithveryseverechronicobstructivepulmonarydisease(COPD)containsignificantlyhighernumbersofalveolarmacrophagesthanthoseofnon-smokersorsmokers.M1andM2macrophagesrepresentpro-andanti-inflammatorypopulations,respectively.However,therolesofM1andM2alveolarmacrophagesinCOPDremainunclear.ImmunohistochemicaltechniqueswereusedtoexamineCD163, CD204 andCD206,asM2markers,expressedonalveolarmacrophagesinthelungsofpatientswithmildtoverysevereCOPD(GlobalInitiativeforChronicObstructiveLungDisease(GOLD)stageI(mild)n = 11,II(moderate)n = 9,III(severe)n = 2,andIV(verysevere)n = 16).Fifteensmokersand10non-smokerswerealsoexaminedforcomparison.ThereweresignificantlyhighernumbersofalveolarmacrophagesinCOPDpatientsthaninsmokersandnon-smokers.ThenumbersandpercentagesofCD163(+), CD204(+)orCD206(+)alveolarmacrophagesinpatientswithCOPDatGOLDstagesIIIandIVweresignificantlyhigherthaninthoseatGOLDstagesIandII,andthoseinsmokersandnon-smokers.InpatientswithCOPD,therewasasignificantnegativecorrelationbetweenthenumberofCD163(+), CD204(+)orCD206(+)alveolarmacrophagesandthepredictedforcedexpiratoryvolumeinonesecond.OverexpressionofCD163, CD204 andCD206onlungalveolarmacrophagesmaybeinvolvedinthepathogenesisofCOPD.KakuYetal.PLoSOne. 2014Jan30;9(1):e87400. doi:10.1371/journal.pone.0087400.eCollection2014.
