APY03isamousemonoclonalantibody27B10thatrecognizesphosphorylatedtyrosineresidues.APY03wasraisedagainstaproprietarymixtureofphosphotyrosinepeptidesconjugatedtoKLH.APY03candetectlessthan10ngofphosphotyrosine-labeledbovineserumalbumin(seeCertificateofAnalysis[COA]).APY03ispurifiedbyproteinGaffinitychromatographyandissuppliedasalyophilizedwhitepowder.EachLotofantibodyisqualitycontrolledtoprovideahighbatchtobatchconsistency.TheLotspecific gpertubecanbefoundintheLotspecificCOAdocuments.APY03-HRPshowshighspecificitytophosphotyrosineproteinsanddoesnotcross-reactwithphosphoserine/threonineproteinsawesternblotassay(Figures1).

A431cellswereeithertreated(+)oruntreated(-)withCalyculinA(CalyA:aserine/threoninephosphataseinhibitor,50nMfor1hour).NIH3T3cellswereeithertreatedoruntreatedwithH2O2-activatedsodiumorthovanadate(Vanadate:aspecifictyrosinephosphataseinhibitor,100 Mfor10minutes).10 gofeachlysatewasresolvedinSDS-PAGEandproteinsweretransferredtoPVDFmembrane.APY03-HRP(1:6000)wasusedtodetecttyrosinephosphorylatedproteins.WesternblotwasdevelopedwithSuperSignalWestDurachemiluminescentreagent(ThermoScientific)andexposuretimewas10seconds.AsshowninFig.1,awiderangeoftyrosinephosphorylatedproteinsweredetectedinNIH3T3cellstreatedwithorthovanadatebutnotinCalyculinAtreatedA431cells.
