Barry McCleary Founder
Prof. Barry McCleary, CEO, received his PhD in 1975 from the University of Sydney, and DScAgr degree in 1989 from the same University. His contributions to analytical chemistry have been recognised at the highest international level, including the Guthrie Medal from the Royal Australian Chemical Institute, the Harvey W. Wiley Award from AOAC International, the Clyde H. Bailey Medal from ICC, the Edith A. Christensen award, and the Applied Research Medal from AACC International.
From 2009-2011, Prof. McCleary served as President-Elect, President and Chair of the Board of AACC International. 简介:
酸水解法和酶法被广泛的应用于淀粉含量测定,酸水解法仅仅适用于纯净的淀粉样品,应用范围有限。酶法的前处理步骤上有各种变化,经过凝胶、液化和糊精化作用,糊精水解生成葡萄糖,zui终以测量的葡萄糖计算淀粉含量。AACC方法76-11特殊之处在于:在高压蒸汽和碱性环境下淀粉糊化成为凝胶,然后再用淀粉葡糖苷酶将淀粉转化为葡萄糖进行测量。AACC方法76-11会低估某些样品的淀粉含量,包括高多糖玉米淀粉和众多的经过加工的谷物产品。今天应用的众多的测定方法都使用了联合处理方法,如在样品处理过程中或直接在淀粉凝胶化后使用耐热α-淀粉酶。对于难凝胶的样品(如高多糖玉米淀粉)使用了氢氧化钠或二甲基亚砜作为溶剂。为确保膳食纤维中的淀粉能够全部溶解,Englyst和Cummings(1988)总结的处理方法中使用了去分支酶,支链淀粉酶。
为了测量极端样品得到满意结果,兼顾数量和可靠性,Megazyme提供了一个总淀粉分析试剂盒,基于使用耐热性淀粉酶和淀粉葡糖苷酶。这一方法已被AOAC和AACC采用 (Method 76.13)。
zui近,耐热淀粉酶可在低pH环境下使用。因此,我们更新了我们的总淀粉测量方法加入了这种酶。这一改进的zui大益处在于可以在同一pH(pH5.0)条件下进行耐热性淀粉酶和淀粉葡糖苷酶孵育,简化步骤以及减少麦芽酮糖(抗耐热性淀粉酶和淀粉葡糖苷酶水解)的产生。另一改进是使用己糖激酶/葡萄糖-6-磷酸脱氢酶/ NADP+在D-葡萄糖处理步骤(K-TSHK5)。Megazyme总淀粉分析方法可以测量广泛范围内食品,饲料,植物和谷类产品(自然的或加工的)。对于大多数样品(例如小麦面粉),淀粉可以在100°C抗耐热性淀粉酶处理步骤中完全可溶。包含高水平抗性淀粉(例如高多糖含量玉米淀粉)样品,需要先用冷2 M KOH 或 热DMSO进行预溶解。含可溶性淀粉和糊精的样品,无需进行抗耐热性淀粉酶处理。
原理:
抗耐热性淀粉酶水解淀粉为可溶性分支麦芽糊精和去分支麦芽糊精。
样品中的抗性淀粉可用2M KOH进行预溶解,再用醋酸钠缓冲液进行中和再进行淀粉酶水解。或者用DMSO在100°C进行溶解。
淀粉葡萄糖苷酶水解麦芽糊精至D-葡萄糖。
D-葡萄糖氧化为D-葡萄糖酸脂,释放出(H2O2),再用过氧化氢酶催化进行显色反应,产生醌亚胺染料。
样品包含高水平的D-葡萄糖和麦芽糊精可以在分析前用80%乙醇进行洗涤。单个样品可在70分钟内完成测定。20个样品可在2小时内完成。
特异性,灵敏度,线性范围和精确性
此试剂盒专用于分析a-葡聚糖(包括淀粉,糖原,植物糖原和非抗性麦芽糊精)。
zui小可区分的吸收值是0.01吸收值范围。这对应于1.0mg的D-葡萄糖(或0.9mg淀粉)/L在zui大样品体积1ml。检测极限:2.0mgD-葡萄糖(或1.8mg淀粉)/L,这对应于0.02吸光度区别在zui大体积1ml中。
此试剂盒在5-100ugD-葡萄糖范围内呈线性。一个样品溶液的重复测定中,吸收值会有0.005至0.010的波动。当样品体积为1ml时,这相当于0.05-1ml/L浓度的D-葡萄糖。如果样品在准备过程中被稀释,结果需要乘以稀释倍数。例如在样品准备时,样品称重,如10g/L,变异范围将在0.02至0.05g/100g。
干扰:
如果D-葡萄糖的转化在5分钟内完成,不会产生干扰。可以通过加入D-葡萄糖至反应完成后试管内(例如50ug在0.1ml)以检测干扰。可以看到吸光度显著上升。
样品内的干扰物质可以通过加入内标进行分析。可以对这标准进行定量校正。样品操作中的损失可以通过在起始步骤中加入D-葡萄糖进行校正。
试剂盒成分
Megazyme总淀粉含量测定试剂盒提供有足够运行100次试验的试剂,并提供有全部的试验方法:
瓶1: 耐热α-淀粉酶(10mL, 3,000U/mL在CERALPHA试剂中pH6.5; 40°C 或1600U/mL在CERALPHA试剂中pH5.0; 40°C)。稳定性>4年, 4°C保存。
瓶2: 淀粉葡糖苷酶(10mL, 3300U/mL在可溶性淀粉)(或200U/mL在p-nitrophenylβ-maltoside*,pH4.5,40°C)稳定性>4年, 4°C保存。

瓶3: GOPOD试剂缓冲液。磷酸钾缓冲液(0.26M,pH7.4),对羟基苯甲酸(0.22M),叠氮化钠(0.4%W/V)稳定性>4年, 4°C保存。
注意:
1. 如果GOPOD缓冲液存储在-20°C,会形成盐结晶,用双蒸水稀释至1L前必须完成溶解。
2. 此溶液含叠氮化钠(0.4%W/V),有毒。
瓶4: GOPOD试剂酶。葡萄糖氧化酶(> 12,000 U) 和过氧化物酶(> 650 U)和4-氨基安替比林(80mg)。冻干粉,稳定性>4年, -20°C保存。
瓶5: 葡萄糖标准溶液(5ml,1mg/mL)在0.2% W/V苯甲酸溶液中。稳定性>4年,室温保存。
瓶6: 标准的玉米淀粉。淀粉含量见标签。稳定性>4年, -20°C保存。
准备试剂溶液
溶液1 用试剂1(100mM醋酸钠缓冲液, pH5.0,自备)稀释1.0mL瓶1成分至30mL 。分成小份后冻存。稳定性>3年, -20°C保存。
注意:如果按照AOAC方法996.11(example b),用试剂4(50mM MOPS缓冲液,pH7.0)稀释酶。
溶液2 此酶不用稀释可以直接使用,由于酶溶液具有一定粘稠度,所以必须使用正确的移液器。4°C下保存,稳定性>3年。
溶液3 用蒸馏水稀释瓶3成分(GOPOD试剂缓冲液)到1L, 立即使用。
溶液4 用20ml溶液3溶解瓶4成分再倒回溶液3瓶子。铝箔纸包裹避光。这就是 葡萄糖测定试剂(GOPOD试剂) 稳定性: 4°C下保存,3个月;-20°C下保存, 12个月;
分装成小份再进行冻存,只能冻融一次。
新鲜配制的试剂呈浅黄色或浅紫色。4°C下保存2-3个月过程中,逐渐变成深紫色。用水为空白对照时,此溶液的吸光度<0.05。
溶液5&6 同瓶5&6
自备试剂
1.醋酸钠缓冲液(100mM, pH5.0) +化钙(5mM)
5.8ml冰醋酸(1.05g/mL)加入900mL蒸馏水,用1M(4g/100mL)NaOH调节pH到5.0,大约需要30mL。4°C下保存2个月.
添加0.74g二水氯化钙完全溶解, 定容到1L, 4°C下保存>6个月。
注意:可通过添加叠氮化钠(0.2g/L)增加此缓冲液的稳定性。室温2年。必须在pH调完后再加入叠氮化钠。酸化的叠氮化钠会释放出有体。
2.醋酸钠缓冲液(1.2M, pH3.8)
69.6ml冰醋酸(1.05g/mL)加入800mL蒸馏水, 用4M NaOH调节pH到3.8, 定容到1L。室温下保存12个月。
3.氢氧化钾溶液(2M)
加入112.2 g KOH 至900 mL 去离子水,搅拌溶解,定容一升。储存于密闭容器。室温下>2年。
4. MOPS缓冲液(50mM,pH7.0)+化钙(5mM) +叠氮化钠(0.02%)(仅用于example b分析样品)
11.55g MOPS(钠盐, Sigma cat. M-9381)加入900mL蒸馏水, 用1M(10%V/V)HCl调节pH到7.0,大约需要17mL。 添加二水氯化钙(0.74g)和叠氮化钠(0.2g), 完全溶解, 然后调整到1L, 4°C下保存6个月。
5. 醋酸钠缓冲液(200mM,pH4.5)+叠氮化钠(0.02%)(仅用于example b分析样品)
冰醋酸(11.8mL,1.05g/mL)加入900mL蒸馏水, 用1M(4g/100mL)NaOH调节pH到4.5,大约需要60mL。加入叠氮化钠(0.2g),完全溶解,然后调整到1L, 4°C下保存6个月。
注意:可通过添加叠氮化钠(0.2g/L)增加此缓冲液的稳定性。室温2年。必须在pH调完后再加入叠氮化钠。酸化的叠氮化钠会释放出有体。

注意事项:
1.GOPOD试剂的孵育时间没有严格规定,但是至少要20分钟,并在60分钟内测定吸光度.
2.每次试验必须设定试剂空白对照和葡萄糖对照(100μg, 四倍)。
a)试剂空白对照: 0.1mL蒸馏水+3.0mLGOPOD溶液
b)葡萄糖对照包括:0.1mL葡萄糖标准溶液(100μg/0.1mL)+3.0mLGOPOD溶液。因子F(页码13和14)通过D-葡萄糖在标准分析中读得的吸光度进行计算(例如100/1.038=96.386)。
3.每次试验必须设定标准面粉或淀粉样品
4.每一新批此的GOPOD溶液,必须验证其与100μg葡萄糖标准反应产生颜色所需要的zui长时间, 通常为15分钟左右。
样品空白:
样品空白按照标准试验程序(example A)进行,将第4步修正为添加3mL蒸馏水,第5步中也用蒸馏水替代其中的淀粉葡糖苷酶.也可以用样品前处理中所用的酒精作为样品空白( example E)。
样品准备示例
A.不含抗性淀粉,D-葡萄糖和麦芽糊精的谷物和食物(建议的程序,pH5.0孵育)。
1.研磨谷物, 过0.5mm筛(35目)。
2.将研磨后样品(准确称量~100mg)加入到试管中(16x120mm),确保全部样品位于试管底部。
3.加入0.2mL乙醇溶液(80%v/v)湿润样品帮助分散, 用漩涡混合器混合。
4.立即加入3mL耐热α-淀粉酶(100mM醋酸钠稀释), 在沸水浴中孵育6分钟(在孵育2分钟,4分钟,6分钟的时强烈振荡试管).
注意:在这一步中强力震荡是关键,目的是确保浆状样品能完全的混匀(去除块状)。同样,每间隔2分钟振荡也是为了防止由于酒精蒸发导致某些样品被喷出试管。如果使用聚丙烯管,增加孵育时间至12分钟,在 4, 8和12强烈震荡。
5.将试管放置于50°C的水浴锅中, 加入0.1mL瓶2成分(淀粉葡糖苷酶,330U在淀粉上)。充分混合,在50°C下孵育30分钟。
6.将全部的溶液转移到100mL的容量瓶中,用洗瓶冲洗试管,并也倒入容量瓶中,然后用蒸馏水调整溶液体积, 充分混匀。取部分溶液离心(3,000rpm,10分钟)。取清澈的未稀释的上清用于分析。
注意:对于含有1-10%淀粉的样品,在第6步中用蒸馏水调整溶液至10mL,充分混匀,离心(3,000rpm,10分钟),该溶液可以直接进行第7步.对于含10-100%淀粉的样品,用蒸馏水稀释1.0mL样品液到10mL再进行第7步。
7.转移两份稀释的样品溶液(0.1mL)到园底试管中(16x100mm)。
8.加入3.0mLGOPOD溶液到每一个试管中(包括葡萄糖对照和试剂空白对照), 然后在50°C下孵育20分钟
9.葡萄糖对照包括0.1mL葡萄糖标准溶液(1mg/mL)+3.0mLGOPOD溶液。 试剂空白对照: 0.1mL蒸馏水+3.0mLGOPOD溶液
10.相对于试剂空白在510nm下测定每一个样品和葡萄糖质控的吸光度。
B.不含抗性淀粉,D-葡萄糖和麦芽糊精的谷物和食物(AOAC 996.11)。
1.研磨谷物, 过0.5mm筛(35目)。
2.将研磨后样品(准确称量~100mg)加入到试管中(16x120mm),确保全部样品位于试管底部。
3.加入0.2mL乙醇溶液(80%v/v),用漩涡混合器混合。
4.立即加入3mL耐热α-淀粉酶(50mM MOPS稀释), 在沸水浴中孵育6分钟(在孵育2分钟,4分钟,6分钟的时强烈振荡试管).
注意:在这一步中强力震荡是关键,目的是确保浆状样品能完全的混匀(去除块状)。同样,每间隔2分钟振荡也是为了防止由于酒精蒸发导致某些样品被喷出试管。如果使用聚丙烯管,增加孵育时间至12分钟,在 4, 8和12强烈震荡。
5.将试管放置于50°C的水浴锅中, 加入醋酸钠缓冲液(4ml,200mM,pH4.5),加入0.1mL瓶2成分(淀粉葡糖苷酶, 20U)。充分混合,在50°C下孵育30分钟。
6. 按照example A 步骤6继续实验。
C.含抗性淀粉,不含D-葡萄糖和麦芽糊精的谷物和食物(建议KOH溶解)
1.研磨谷物, 过0.5mm筛(35目)。
2.将研磨后样品(准确称量~100mg)加入到试管中(16x120mm),确保全部样品位于试管底部。
3.加入0.2mL乙醇溶液(80%v/v),用漩涡混合器混合。
4.放入搅拌架子,加入2ml 2M KOH至每个试管中,冰水混合浴中搅拌20分钟左右,重悬粉末和溶解抗性淀粉。
注意:
1.不要使用涡旋仪混匀,会使淀粉乳化。
2.确保加入KOH时,试管处于剧烈震荡状态。这是为了避免淀粉形成团块难以溶解。
5.当试管在磁力搅拌器上搅拌时,加入8ml 1.2M醋酸钠缓冲液(pH3.8)至每个试管。立即加入0.1ml耐热α-淀粉酶(瓶1)和0.1mL淀粉葡糖苷酶(瓶2),充分混合,在50°C下孵育
6. 孵育30分钟,间断混匀。
7. 样品淀粉含量>10%:将全部的溶液转移到100mL的容量瓶中,用洗瓶冲洗试管,并也倒入容量瓶中,然后用蒸馏水调整溶液体积至100ml, 充分混匀。取部分溶液离心(1800g,10分钟)。
8. 样品淀粉含量<10%:直接离心(1800g,10分钟)。总体积约10.4ml(此体积可能有一定波动,特别是湿样品用于分析时,估计体积用于计算)。
9. 按照example A 步骤7继续实验。
D.含抗性淀粉,不含D-葡萄糖和麦芽糊精的谷物和食物(AOAC 996.11 DMSO溶解)
1.研磨谷物, 过0.5mm筛(35目)。
2.将研磨后样品(准确称量~100mg)加入到试管中(16x120mm),确保全部样品位于试管底部。
3.加入0.2mL乙醇溶液(80%v/v),用漩涡混合器混合。
4.立即加入2ml DMSO,涡旋混匀。沸水浴5分钟。
5.按照example A 步骤4继续实验。
E.含D-葡萄糖和麦芽糊精的谷物和食物
1.研磨谷物, 过0.5mm筛(35目)。
2.将研磨后样品(准确称量~100mg)加入到试管中(16x120mm),确保全部样品位于试管底部。
3.加入5mL乙醇溶液(80%v/v),80-85°C孵育5分钟。涡旋混匀,再加入5ml乙醇溶液(80%v/v)。
4.离心1800g(3000rpm)10分钟。去除上清。
5.10mL乙醇溶液(80%v/v)重悬沉淀。离心去除上清。
6. 按照example A 步骤4继续实验。
若样品含抗性淀粉,按照example C步骤4继续实验。

The Total Starch (AA/AMG) test kit is used for the measurement and analysis of total starch in cereal flours and food products. This kit now contains an improved α-amylase that allows the amylase incubations to be performed at pH 5.0 (as well as pH 7.0).
Colourimetric method for the determination of Total Starch in
cereal products, feeds, foodstuffs and other materials
Principle:
(α-amylase, 100°C ± DMSO)
(1) Starch granules + H2O → maltodextrins
(amyloglucosidase)
(2) Maltodextrins + H2O → D-glucose
(glucose oxidase)
(3) D-Glucose + H2O + O2 → D-gluconate + H2O2
(peroxidase)
(4) 2H2O2 + p-hydroxybenzoic acid + 4-aminoantipyrine →
quinoneimine + 4H2O

Detection limit: 1-100% of sample weight
