The T7 Gp4A Helicase exhibits a 5’ to 3’ helicase activity as well as primase activity. It is highly processive, traveling an average of 75 kb of single stranded DNA before dissociating.

Purified from an E. coli strain that overexpresses the gp4A gene isolated from the T7 phage.            

Reaction condition:

   

1X Gp4A Reaction Buffer:
35 mM Tris–acetate, pH 7.5
11 mM magnesium acetate
5 mM DTT
0.01% Triton X-100

Supplement with 2 mM dTTP.
Incubate at 25 - 37ºC.

NOTE:
The concentration of dTTP should be optimized for each specific application.
For the primase activity, supplement reactions with NTPs.
