Description:

RecombinanthumansolubleFGFR-4wasfusedwiththeFc-partofhumanIgG1.HumanrecombinantsolubleFGFR-4/Fcisadisulfide-linkedheterodimericprotein.InthereducedformtheglycosylatedsubunitsofsFGFR-4alpha/humanFcchimeradisplayamolecularmassof80-85kDa.Fibroblastgrowthfactors(FGFs)compriseafamilyofatleasteighteenstructurallyrelatedproteinsthatareinvolvedinamultitudeofphysiologicalandpathologicalcellularprocesses,includingcellgrowth,differentiation,angiogenesis,woundhealingandtumorigenesis.ThebiologicalactivitiesoftheFGFsaremediatedbyafamilyoftypeItransmembranetyrosinekinaseswhichundergodimerizationandautophosphorylationafterligandbinding.FourdistinctgenesencodingcloselyrelatedFGFreceptors,FGFR1-4,areknown.AllfourgenesforFGFRsencodeproteinswithanN-terminalsignalpeptide,threeimmunoglobulin(Ig)-likedomains,anacid-boxregioncontainingarunofacidicresiduesbetweentheIgIandIgIIdomains,atransmembranedomainandthesplittyrosine-kinasedomain.MultipleformsofFGFR-1to-3aregeneratedbyalternativesplicingofthemRNAs.AfrequentsplicingeventinvolvingFGFR-1and-2resultsinreceptorscontainingallthreeIgdomains,referredtoasthealphaisoform,oronlyIgIIandIgIII,referredtoasthebetaisoform.OnlythealphaisoformhasbeenidentifiedforFGFR-3andFGFR-4.AdditionalsplicingeventsforFGFR-1to-3,involvingtheC-terminalhalfoftheIgIIIdomainencodedbytwomutuallyexclusivealternativeexons,generateFGFreceptorswithalternativeIgIIIdomains(IIIbandIIIc).AIIIaisoformwhichisasecretedFGFbindingproteincontainingonlytheN-terminalhalfoftheIgIIIdomainplussomeintronsequenceshasalsobeenreportedforFGFR-1.MutationsinFGFR-1-3havebeenfoundinpatientswithbirthdefectsinvolvingcraniosynostosis.ThecomplexpatternsofexpressionofthesereceptorsaswellasthespecificityoftheirinteractionswiththevariousFGFligandfamilymembersareunderinvestigation.
