SMCCcrosslinker(Succinimidyl-4-[N-maleimidomethyl]cyclohexane-1-carboxylate)isaheterobifunctionalproteincrosslinker.SMCCconjugationoccursviaamaleimidegroupthatissulfhydryl(thiol;-SH)reactiveandaNHSestergroupthatisaminereactiveandformsstable,covalentproteincrosslinks.SMCCcrosslinkermustbedissolvedinanorganicsolvent,suchasDMSOorDMF,andthencanbeaddedtoanaqueouscrosslinkingreaction.SMCCcrosslinkingreagentispermeableacrossthelipidbilayerofthecellmembraneandcanbeusedtocrosslinkintracellularproteinsandpeptides.

SMCCisachoicereagentforproducingstablemaleimideactivatedcarrierproteinsthatwillreactwithsulfhydryls(thiols).Thecyclohexaneringinthe9-atom,non-cleavablespacerarmofSMCCdecreasestherateofhydrolysisofthemaleimidegroup.ThusallowingproteinswhichhavebeenmaleimideactivatedwithSMCCtobelyophilized,stored,andlaterconjugatedtosulfhydrylcontainingmolecules.Numerousmaleimideactivatedproteinproductsareproducedinthismannerbyfirstformingthecrosslinkwithprimaryamines,asfoundinlysineresidues,onthecarrierproteinviatheNHS-esteroftheSMCCcrosslinkingreagentandconvertingthemtoreactivemaleimides.Therefore,themaleimideactivatedintermediateisquitestable,maybelyophilized,andstoredforlaterconjugationtoahaptenforantibodyproduction.

SATAProteinModifiercanbeusedtoaddsulfhydrylgroups(thiols,-SH)toproteinsorpeptidespriortoSMCCcrosslinking.
