Inositolhexakisphosphatekinase2(IP6K2)catalyzestheproductionofdiphosphoinositolpentakisphosphate(IP7)andhasaroleasaproapoptoticgenesensitizingcancercellstoapoptosisbycellstressorsandanticancerdrugsallofwhichdependonIP6K2catalyticactivity.IP6K2islocatedinthe3p21.31chromosomalregion,whichoftenundergoesallelelossinavarietyofhumancancers.Someheatshockproteins,especiallyHsp90,canbeantiapoptoticandthetargetsofanticancerdrugs.Hsp90bindsIP6K2andinhibitsitscatalyticactivity.DrugsandselectivemutationsthatabolishHSP90-IP6K2bindingelicitactivationofIP6K2,leadingtocelldeath.Thus,theprosurvivalactionsofHSP90reflecttheinhibitionofIP6K2,suggestingthatselectivelyblockingthisinteractioncouldprovideeffectiveandsafermodesofchemotherapy.
