Theantibodyhassignificantbiotin-avidinHRPstainingata1/300-1/500dilutioninratthalamus,hippocampusandcortex.Optimaldilutionwillvarydependinguponfixation,labelingtechniqueand/ordetectionsystem;therefore,adilutionseriesisrecommended. Theantibodyisspecificforthe~125kmonomerandthe~250kmGluR1adimerinWesternblotsofratbrainextracts. ImmunolabelingformGluR1aiscompletelyabolishedbypreadsorptionwithsyntheticratmGluR1a(1116-1130)at5µgpermLofdilutedantibody. Note: Ifneeded,lowlevelsofglutaraldehyde(0.1-0.3%)maybeused  inconjunctionwithparaformaldehyde.

PhotoDescription:IHCimageofneuronsstainingforthemGluR1alphaintherathippocampus(above)andwesternblotshowingspecificityoftheantibodyat132kDa(below).ForIHC,thetissuewasfixedwith4%formaldehydein0.1Mphosphatebuffer,beforebeingremovedandpreparedforvibratomesectioning.FloatingsectionswereincubatedatRTin10%goatseruminPBS,beforestandardIHCprocedure.Primaryantibodywasincubatedat1:500for48hours,goatanti-rabbitsecondarywassubsequentlyaddedfor1hourafterwashingwithPBS.Lightmicroscopystainingwasachievedwithstandardbiotin-streptavidin/HRPprocedureandDABchromogen.
