Themonoclonalantibody6E9recognizesmousemembrane-boundaswellassolubletriggeringreceptorexpressedonmyeloidcells-2(TREM-2).TREM-2isa26kDatransmembraneglycoproteinthatconsistsofasingleextracellularimmunoglobulin-likedomain,atransmembraneregionwithachargedlysineresidueandashortcytoplasmictail.ItassociateswithDNAX-activationprotein12(DAP12)forsignalingandfunction.TREM-2isexpressedonimmaturemonocyte-deriveddendriticcells.Afteractivationbymicrobialproductsortumornecrosisfactor(TNF)andTNF-relatedproteins,dendriticcellsdownregulatetheexpressionofTREM-2.TREM-2isalsoexpressedbyosteoclastsandmicroglia,whereitisinvolvedinbonemodelingandbrainfunction,respectively.AnotherroleofTREM-2mightbepromotingtheremovalofapoptoticcells,organicmatrixandmacromoleculesbymicroglia.DefectsinTREM-2areacauseofpolycysticlipomembranousosteodysplasiawithsclerosingleukoencephalopathy(PLOSL),alsocalledpreseniledementiawithbonecystsorNasu-Hakoladisease(NHD).TREM-2,likeTREM-1,canbecleavedonthemembranetoreleaseasolubleformofTREM-2(sTREM-2).ElevatedlevelsofsTREM-2inCSFofmultiplesclerosispatientshavebeendetected.Thiselevatedlevelmayinhibittheanti-inflammatoryfunctionofthemembrane-boundreceptorsuggestingsTREM-2tobeapossIBLetargetforfuturetherapies.
