Themonoclonalantibody6B1recognizeshumantriggeringreceptorexpressedonmyeloidcells1(TREM-1).TREM-1isatransmembraneglycoproteinbelongingtoafamilyrelatedtothenaturalkillercellreceptors.TREM-1isanimportantactivatingreceptor,of26kDa,involvedintheinnateinflammatoryresponseandinsepsis.TREM-1isexpressedatlowlevelsintheearlydevelopmentofthehematopoieticsystemandinthepromonoisticstageandathighlevelsonneutrophilgranulocytes(PMN),monocytes,andmacrophagesubsets.TheexpressionofTREM-1isupregulatedbymicrobialproducts,thatis,bytoll-likereceptorligandssuchaslipoteichoicacid(LTA)ofGram-positiveorlipopolysaccharide(LPS)ofGram-negativebacteria.LigationofTREM-1issynergisticwithTLRagonistsontheactivationofreceptorbearingcells.PlateletsexpressanaturalligandforTREM-1.Receptorligationactivatesthefullrepertoireofeffectorfunctions.TREM-1isalsoproducedinasolubleform(sTREM-1)of17kDawhichisreleasedinhumansafterendotoxinexpositionorinpatientssufferingfromseverepneumoniaorsepsis.sTREM-1canbemeasuredinbiologicalfluidsandmaybeusefulasadiagnostictool.Themonoclonalantibody6B1recognizesbothmembraneboundandsolubleTREM-1.Also,themonoclonalantibody6B1neutralizestheinteractionbetweenTREM-1anditsligand.
