本研究通过应用脂质体剂型包裹抗代谢药物注入晶体囊外摘除术后的晶体囊袋内,观察药物在眼内的释放及对晶体上皮细胞的作用,探索药物防治后发性白内障的可能性。结果显示:(1)采用脂质体剂型包裹高三尖杉酯碱(HH)、氟尿嘧啶(FU)、阿霉素(ADM)、地塞米松(DXT)可以在眼内缓慢释放;(2)眼内缓慢释放时间为2周;对晶体上皮细胞作用显效时间达4周:光镜下表明FU、HH、ADM组能抑制残留晶体上皮细胞的增殖;电镜下可见细胞浆空泡变性、线粒体肿胀等形态改变,与对照组、DXT组间有显著性差异(P<0.01);(3)所有脂质体剂型药物组与对照组在手术后对角膜厚度、内皮细胞的数量和形态的影响均无显著性差异(P>0.05)。

Purpose:Toinvestigatethemethodofdeterminationtheconcentrationofdaunoru-bicine(DNR)andliposomeencapsulateddaunorubicine(LDNR)ofaqueoushumorinrabbiteyes.

目的:探讨药物柔红霉素(daunorubicine,DNR)及其脂质体在房水中维持的浓度及时间。方法:20只兔眼于晶体囊外摘除术中分别注入0.1ml之1mg/mlDNR及其脂质体(LDNR),术后12、24、48小时和1周时间采集房水,应用高效液相色谱法测定房水中DNR的浓度。结果:术后12、24、48小时注入DNR和LDNR眼均可测出DNR,且LDNR眼浓度明显高于DNR眼,术后1周LDNR眼仍可测出DNR,但注入DNR眼无法测出。结论:应用高效液相色谱法可测定兔房水中的DNR浓度,且LDNR眼浓度明显高于DNR眼,有明显的缓释延时效应。眼科学报1998;14:232~235。

目的探讨骆驼蓬总碱(TAH)及其脂质体(LTAH)对兔后囊混浊(PCO)形成的影响。方法用改良冻融法制备LTAH。10只健康纯种新西兰白兔分别每眼行晶状体囊外摘出术并分别注入01ml浓度为02mg/mlTAH及LTAH。观察术后后囊、手术前后眼压、角膜内皮细胞、视网膜电图及组织病理学变化。结果注药术后4周实验眼与对照眼角膜内皮细胞密度及形态、视网膜电图的b波振幅及潜时无明显差别。组织病理学检查显示对照组眼明显残留晶状体上皮细胞(LEC)增生并有Soemmering环形成,而用药眼无明显的残留LEC增殖且细胞空泡变性。结论TAH及LTAH能抑制兔眼残留LEC增殖,有可能成为一种预防后发性白内障的药物。
