| Scientific Background: | A series of studies in Caenorhabditis elegans, Schizosaccharomyces pombe and mammals has shed light on the important roles that cullin-based ubiquitin ligases play in the regulation of diverse developmental processes and environmental responses in eukaryotic organisms.The ubiquitin-like protein RUB1 (NEDD8 in mammals) is conjugated to target proteins by a mechanism similar to that of ubiquitin conjugation in Arabidopsis. It has been shown that the Skp1-Cul1/Cdc53-F-box (SCF) subunit CUL1 is modified by RUB1 in vivo. The formation of RUB-AtCUL1 is dependent on AXR1 and ECR1 and expression of these proteins is restricted to zones of active cell division and cell elongation, consistent with their role in growth regulation. These results provide strong support for a model in which RUB conjugation of AtCUL1 affects the function of SCF E3s. |
