脂质体或许大家更熟悉,它的历史已超过20年,是最受欢迎的体外导入载体。脂质体结合或包围核酸,进而被细胞吸收。Invitrogen公司的脂质体Lipofectin、Lipofectamine、Lipofectamine 2000已畅销十余年。脂质体还是携带小分子RNA进入细胞的有力武器。Ambion(现属于生命科技公司)就开发出siRNA转染试剂-siPORT NeoFX,Invitrogen也修改了阳离子脂质体的配方,专门为小分子RNA推出RNAiMAX。
当然,转染不仅仅限于细胞系,许多研究人员将他们的目光投向体内转染。一些生物公司如Polyplus-transfection、Altogen Biosystems等利用脂质体或可生物降解的聚合物作为载体,将DNA片段或siRNA导入体内。然而,它们都不是理想的载体。脂质体的导入效果虽比聚合物要持久,但它可能引起更多的免疫反应,这也不是研究人员所期望的。也正是这些不足,导致了近几年纳米颗粒的出现。2007年,Sigma-Aldrich推出了N-TER纳米颗粒siRNA转染试剂,让siRNA结合在纳米颗粒的表面,通过N-TER肽段导入体内。
在RNAi研究领域,越来越多的研究人员开始对高通量的筛选感兴趣。瑞典Cellectricon公司也注意到这一点,他们认识到转染是RNAi筛选中的主要瓶颈之一,急需一种高通量的转染方法。于是他们花了两年的时间,开发出第一台全自动的转染系统-Cellaxess HT电穿孔仪。这台仪器能够自动移液,自动在细胞中加入siRNA,自动将移液吸头转换成电穿孔元件,并进行电穿孔,最后加入细胞培养基进行复苏。Cellaxess HT每天能转染50000个孔。蚂蚁淘之前介绍过的Nucleofector核转染系统也是类似的电穿孔仪器,它能够对96孔板进行转染。
说到底,转染的目标其实很简单,就是用最少量的DNA或RNA去转染最多的细胞。当然,还要尽量减少或避免对细胞的毒性、脱靶效应和免疫反应。别看这个目标简单,实现起来还真不容易。无论是试剂还是仪器,都有需要改进的地方,同时,新技术也在不断萌芽。也许在不久的将来,转染将不再是实验中的瓶颈,轻轻松松就能实现。
