人溶酶体磷脂酶A2（LPLA2）负责正常的脂质代谢，并且与其他已知的PLA2不同，因为LPLA2仅在酸性环境（pH4.5左右）中有活性。最近的研究表明LPLA2与药物诱导的磷脂质过多症（DIPL）有关。DIPL是长期使用某些阳离子两亲药物（如氟西汀（Prozac™，Sarafem）和胺碘酮）引发的细胞内磷脂过度积累的一种疾病。DIPL通过组织炎症和器官损伤进展。如果没有及时发现，DIPL可能导致死亡。FDA已确定DIPL是一个严重的药物安全问题。

Echelon的LPLA2活性测定（K-7000A）是一种同源测定，旨在检测LPLA2生物和纯化来源的LPLA2活性。该测定使用猝灭的底物，其在被LPLA2水解时发荧光。淬灭的底物的sn-1位置被酰胺基保护，以消除磷脂酶A1的非特异性转换。酸性测定条件进一步改善LPLA2对水解猝灭底物的选择性。这种直接的生物化学方法在稳健且易于使用的体外基于板的测定中提供了特定的定量测量，用于快速测量LPLA2活性。
