Luciferinisacommonbioluminescentreporterusedforinvivoimagingoftheexpressionoftheluciferase.ThissubstrateforthefireflyluciferaseenzymeutilizesATPandMg²⁺ascofactorsandemitsacharacteristicyellow-greenemissioninthepresenceofoxygen,whichshiftstoredlightinvivoat37°C.

Luciferinisacommonreagentusedthroughoutthebiotechnologyfieldandspecificallyforinvivoimaging.Luciferaselabeledtumorcells,stemcellsorinfectiousdiseasesareofteninoculatedintoresearchanimalssuchasratsormiceforinvestigation.Theinjectionofluciferinallowsforthereal-time,noninvasivemonitoringofdiseaseprogressionand/ordrugefficacyinthesemodelsystemsthroughBioluminescenceImaging(BLI).

DMNPE-cagedluciferincanbeusedtomeasureintracellularfunctionssinceitreadilycrossescellmembranes.Onceitisinsidethecell,activeluciferincanbereleasedbyUVlightorbytheactionofendogenousintracellularesterases.DMNPE-cagedluciferinmakesiteasiertofollowthechangesingeneexpressioninlivecells.ItcanalsobeusedincombinationwithDMNPE-cagedATP.
