Description
ThethreemammalianisoformsofTGF-β,TGF-β1,β2,andβ3,signalthroughthesamereceptorandelicitsimilarBIOLOGicalresponses.Theyaremultifunctionalcytokinesthatregulatecellproliferation,growth,differentiationandmotility,aswellassynthesisanddepositionoftheextracellularmatrix.Theyareinvolvedinvariousphysiologicalprocesses,includingembryogenesis,tissueremodelingandwoundhealing.Theyaresecretedpredominantlyaslatentcomplexes,whicharestoredatthecellsurfaceandintheextracellularmatrix.ThereleaseofbiologicallyactiveTGF-βisoformfromalatentcomplexinvolvesproteolyticprocessingofthecomplexand/orinductionofconformationalchangesbyproteinssuchasthrombospondin-1. TGF-β1isthemostabundantisoformsecretedbyalmosteverycelltype.ItwasoriginallyidentifiedforitsABIlitytoinducephenotypictransformationoffibroblasts,andrecentlyithasbeenimplicatedintheformationofskintumors.RecombinantHumanTGF-β1derivedfromHEK293cellsisa25.0 kDaproteinwitheachsubunitcontaining112 aminoacidresidues,linkedbyasingledisulfide bond.
