BMX(BonemarrowXkinase)isacytoplasmictyrosinekinaseidentifiedbyreversetranscriptionofmRNAisolatedfromhumanbonemarrowandmappedtothechromosomalbandXp22.2.Thefulllengthproteinis675aminoacidswithatyrosinekinasedomain,anaminoterminalpleckstrindomain,aswellasanSH3andSH2domain.DirectcomparisonofBMXsprimarysequencewithotherkinasesshowedthatthisishighlyrelatedtothefamilyofBTK/ITK/TEC.BMXkinaseisexpressedinfetalandadulttissues,withthehighestexpressioninheart,testis,smallintestineandcolon.Itisundetectableinspleen,brain,kidney,andpancreas.FurtheranalysisofmRNAexpressionshowedthatBMXisexpressedinhematopoietictissuesandneutrophilicgranulocytes,andinpatientswithacuteandmyeloidleukemia.ThelevelsofBMXmRNAweresubstantiallylowerinpatientswithacuteandchroniclymphoidleukemias,thussuggestingthatBMXmaybeimportantduringmyelopoiesis.Itisexpressedinavarietyofhematopoietic,epithelialandendothelialcells.
