Schwanncellmedium(SCM)isacompletemediumdesignedforoptimalgrowthofnormalhumanSchwanncellsinvitro.Itisasterile,liquidmediumwhichcontainsessentialandnon-essentialaminoacids,vitamins,organicandinorganiccompounds,hormones,growthfactors,tracemineralsandalowconcentrationoffetalbovineserum(5%).ThemediumisbicarbonatebufferedandhasapHof7.4whenequilibratedinanincubatorwithanatmosphereof5%CO2/95%air.Themediumisformulated(quantitativelyandqualitatively)toprovideadefinedandoptimallybalancednutritionalenvironmentthatselectivelypromotesgrowthofnormalhumanastrocytesinvitro.

ThawSCGS,FBSandP/Ssolutionat37oC.GentlytilttheSCGStubeseveraltimesduringthawingtohelpthecontentsdissolve.Makesurethecontentsofthesupplementarecompletelydissolvedintosolutionbeforeaddingtothemedium.Rinsethebottleandtubeswith70%ethanol,andthenwipetoremoveexcess.Removethecap,beingcarefulnottotouchtheinteriorthreadswithfingers.AddSCGS,FBSandP/Ssolutionintobasalmediuminasterilefield,mixwellandthenthereconstitutedmediumisreadyforuse.Sinceseveralcomponentsofthismediumarelight-lABIle,itisrecommendedthatthemediumnotbeexposedtolightforlengthyperiodsoftime.Ifthemediumiswarmedpriortouse,donotexceed37oC.Whenstoredinthedarkat4oC,thereconstitutedmediumisstableforonemonth.
